Background
This product is a research-grade biosimilar of Bectumomab, offered as an unconjugated, non-therapeutic analog of the originator molecule for research use only. It is not a clinical drug and is supplied without any radiolabel or conjugate. Bectumomab is the murine LL2 Fab' fragment directed against human CD22; the originator was developed as a technetium-99m-labeled imaging agent (marketed as LymphoScan) for radioimmunoscintigraphy of B-cell non-Hodgkin lymphoma. Consistent with that heritage, this reagent is built around the CD22 target and provided as a Fab' fragment of murine IgG2a, kappa framework. As a biosimilar analog it is intended to reproduce the binding specificity of the originator so investigators can study CD22 engagement, benchmark detection assays, and use it as a reference or control reagent. It is manufactured to research standards and is well suited to in-vitro and functional characterisation work rather than administration as a therapeutic. Because the target is human CD22, the reagent is primarily applied in human-cell and biochemical systems. CD22 (Siglec-2; UniProt P20273) is a B-cell-restricted transmembrane sialoglycoprotein of the immunoglobulin superfamily and the Siglec family, recognising alpha-2,6-linked sialic acid ligands. It functions as an inhibitory co-receptor of the B-cell receptor, with cytoplasmic ITIM motifs that recruit SHP-1 to dampen BCR signalling and set thresholds for B-cell activation, thereby contributing to peripheral tolerance and homeostasis. Expression is largely confined to the B-cell lineage, appearing in the cytoplasm of pro- and pre-B cells and on the surface of mature B cells, while typically lost on plasma cells. Its restricted expression and rapid internalisation upon antibody binding make CD22 an attractive target in B-cell malignancies, where it is present on many non-Hodgkin lymphomas and leukemias, and a focus for imaging, immunotoxin, and antibody-based approaches.
