This product is an unconjugated, non-therapeutic recombinant analog of urelumab, a fully human agonist antibody directed against 4-1BB (CD137, TNFRSF9). It is manufactured for research use only and is not the clinical drug; it is not intended for human or veterinary use. The reagent reproduces the antigen-binding specificity of the originator against the human 4-1BB target, making it useful as a defined tool for interrogating 4-1BB costimulatory signaling, as a positive control or benchmarking reagent in binding and functional screens, and in the development and characterization of downstream biologics (for example agonist-antibody engineering, ADCC/ADC workflows, and comparator studies). It supplied unconjugated so users can apply their own detection or conjugation chemistry. Research-grade material is offered at low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options) and in bulk milligram-to-gram quantities suitable for in-vitro assays, cell-based systems, and preclinical model work. Because it is built around the human 4-1BB target rather than the formulated therapeutic, it should be treated as a characterization and control reagent, not a substitute for the licensed product. 4-1BB (CD137, encoded by TNFRSF9) is a member of the tumor necrosis factor receptor superfamily and functions as an inducible costimulatory receptor. It is largely absent on resting T cells and is upregulated following activation, and it is also expressed on activated NK and NKT cells, regulatory T cells, dendritic cells, monocytes, and other myeloid populations. Engagement by its natural ligand 4-1BBL (CD137L, TNFSF9) drives receptor trimerization and higher-order clustering, recruiting TRAF1/TRAF2 adaptors and activating NF-kB and MAPK signaling. The resulting costimulation promotes T cell proliferation, survival (anti-apoptotic signaling), cytokine secretion such as IL-2 and IFN-gamma, and enhanced cytotoxic effector function, particularly in CD8+ T cells. Because of this activity, 4-1BB is a well-studied target for cancer immunotherapy and is a common costimulatory domain in engineered receptor constructs.
