Background
This reagent is a fully mouse bispecific antibody that simultaneously engages mouse CD19 (clone 1D3) and mouse CD3 epsilon (clone 145-2C11), formatted as an IgG2a, kappa backbone carrying LALA-PG Fc mutations to abolish FcgammaR binding and complement activation. By cross-linking CD3 on T cells to CD19 on B cells, it redirects polyclonal T-cell cytotoxicity against the B-lineage compartment independently of TCR specificity, functioning as a mouse surrogate for CD19-directed T-cell engagers. The all-mouse sequence and silenced Fc make it well suited to immunocompetent syngeneic models, where it avoids anti-drug responses and Fc-mediated confounders, allowing the engager mechanism to be studied in isolation. It is manufactured to low endotoxin and supplied in bulk (mg to gram) research-grade format for in-vivo dosing, pharmacology, and immune-monitoring workflows. As a research-use-only reagent it is intended for in-vivo functional studies of T-cell redirection, B-cell depletion, cytokine release, and T-cell engager pharmacodynamics rather than diagnostic or therapeutic use. CD19 is a B-lineage transmembrane glycoprotein expressed from the pro-B stage through mature B cells but lost on terminally differentiated plasma cells. It forms a co-receptor complex with CD21 and CD81 that lowers the threshold for B-cell receptor signalling, making it central to B-cell activation and a stable, near-pan-B-cell target for immunotherapy. CD3 epsilon is an invariant signalling subunit of the CD3 complex that associates with the T-cell receptor and the CD3 gamma, delta and zeta chains to transduce antigen-recognition signals into T-cell activation. Because CD3 epsilon is present on essentially all mature T cells regardless of antigen specificity, antibodies to it can trigger TCR-independent T-cell activation. A bispecific that simultaneously binds CD19 and CD3 epsilon forms an immunological synapse that redirects any nearby T cell to kill the CD19-positive B cell it is tethered to.
