Background
This product is an unconjugated, non-therapeutic recombinant analog of pacibekitug, an antibody directed against human interleukin-6 (IL-6). It is supplied for research use only and is not the clinical drug, nor is it intended for human or veterinary use. Built around the IL-6 target with a human IgG2-kappa framework, it is intended as a tractable reagent for studying IL-6 biology and IL-6-directed antibody workflows in vitro and in preclinical settings. Typical applications include use as a binding and neutralization reagent, an isotype-matched comparator or positive control in assay development, a benchmarking tool for characterizing anti-IL-6 candidates, and a building block for ADCC or ADC development studies. It is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options), and can be provided in bulk milligram-to-gram quantities to support scale-up, assay standardization, and reproducibility across experiments. Because it recognizes a human target, it is most suited to human-cell and human-protein systems rather than mouse in-vivo dosing. Interleukin-6 (IL-6; UniProt P05231) is a pleiotropic cytokine central to inflammation, immune regulation, hematopoiesis, and the acute-phase response. It is produced by many cell types, including macrophages, T cells, fibroblasts, and endothelial cells, in response to infection and tissue injury. IL-6 signals through a receptor complex comprising the IL-6 receptor alpha (IL-6R, CD126) and the signal-transducing subunit gp130 (IL-6ST, CD130). Classic signaling uses membrane-bound IL-6R, while trans-signaling via soluble IL-6R broadens responsiveness to cells lacking membrane IL-6R. Downstream activation of JAK/STAT3, alongside RAS/MAPK and PI3K pathways, drives acute-phase protein synthesis, B-cell differentiation, and Th17 responses. Dysregulated IL-6 is implicated in chronic inflammatory and autoimmune disease, cytokine release syndrome, and tumor biology, making it a well-validated therapeutic target.
