Background
This product is an unconjugated, non-therapeutic recombinant analog of manfidokimab, a fully human IgG4-kappa monoclonal antibody directed against the human interleukin-4 receptor alpha subunit (IL4R / IL-4Ralpha, CD124). It is manufactured for research use only and reproduces the antigen-binding specificity of the originator in a format suitable for laboratory work; it is not the clinical drug and is not intended for human or veterinary use. Because IL-4Ralpha is the shared signalling chain of both the type I and type II receptors, an IL-4Ralpha-directed antibody is a convenient single reagent for simultaneously probing IL-4 and IL-13 biology. Typical applications include use as a positive control or benchmark in binding and neutralisation assays, as a capture or detection reagent, as a tool for characterising IL-4Ralpha expression, and in antibody-engineering or biosimilarity workflows. The antibody is offered as low-endotoxin, carrier-free material (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and is available in bulk milligram-to-gram quantities to support in-vitro assay development and preclinical model standardisation. Interleukin-4 receptor subunit alpha (IL-4Ralpha, CD124; UniProt P24394) is the high-affinity ligand-binding chain shared across IL-4 and IL-13 signalling. In the type I receptor, IL-4Ralpha pairs with the common gamma chain (gamma-c) and responds to IL-4, predominantly on haematopoietic cells. In the type II receptor, IL-4Ralpha pairs with IL-13Ralpha1 and responds to both IL-4 and IL-13, chiefly on non-haematopoietic and myeloid cells. Ligand engagement activates JAK1/JAK3 (type I) or JAK1/JAK2/TYK2 (type II) and drives STAT6 phosphorylation, with IRS-2/PI3K contributions. This axis governs Th2 differentiation, IgE class switching, goblet-cell and mucus responses, and M2 macrophage polarisation, making IL-4Ralpha a central node in type 2 (allergic) inflammation and fibrosis.
