Background
This product is an unconjugated, non-therapeutic recombinant research analog of elarekibep, an engineered protein antagonist directed against human interleukin-4 receptor alpha (IL-4Ralpha, IL4R; UniProt P24394). The originator elarekibep (also referred to in the literature as PRS-060 / AZD1402) is an inhaled Anticalin-class binder developed as a high-affinity, slowly dissociating antagonist of IL-4Ralpha for type 2 inflammatory disease. Because IL-4Ralpha is the shared receptor subunit used by both IL-4 and IL-13, a single IL-4Ralpha-blocking molecule can interrupt signalling driven by both cytokines. The research-grade analog reproduces the target-binding activity of the originator and is intended purely for laboratory use (RUO); it is not the clinical product and is not for human or veterinary use. It is supplied unconjugated and, depending on grade, as low-endotoxin material (research grade typically less than 1 EU/mg; ultra-low grade less than 0.5 EU/mg), suitable as an in-vivo-standard-format reagent and available in bulk milligram-to-gram quantities for assay development, receptor-blockade studies, screening cascades, and preclinical characterisation work. Interleukin-4 receptor alpha (IL-4Ralpha, gene IL4R; UniProt P24394) is a type I transmembrane glycoprotein and the central ligand-binding subunit of the IL-4/IL-13 signalling system. It binds IL-4 with high affinity and then pairs either with the common gamma chain (type I receptor, mainly on hematopoietic cells) or with IL-13Ralpha1 (type II receptor, on many non-hematopoietic cells); the type II complex is also engaged by IL-13. Receptor assembly activates JAK1/JAK3 (or JAK1/TYK2/JAK2) and drives STAT6 phosphorylation, promoting Th2 differentiation, IgE class switching, goblet-cell and mucus responses, and alternative macrophage activation. IL-4Ralpha therefore sits at the convergence point of type 2 (allergic) inflammation, making it a validated node for asthma, atopic dermatitis, and related conditions. A soluble ectodomain form also exists.
