Background
This product is an unconjugated, non-therapeutic recombinant analog of nisevokitug (also known in the literature as NIS-793), a human IgG2-lambda monoclonal antibody built around the TGF-beta 1 (TGFB1) target. It is supplied for research use only (RUO) and is not the clinical drug; it is not intended for human or veterinary use. The analog is designed to recapitulate the target-binding behavior of the originator against TGF-beta and is useful as a tool reagent for in-vitro and preclinical work: as a positive control or reference binder in TGF-beta pathway assays, for neutralization and ligand-blocking experiments, for benchmarking against other anti-TGF-beta reagents, and as a well-defined molecule in assay development and ADCC/ADC-development workflows. It is offered unconjugated in bulk quantities (milligram to gram scale) and in low-endotoxin research grade (<1 EU/mg) and ultra-low-endotoxin (<0.5 EU/mg) formats suitable for endotoxin-sensitive cell-based and preclinical model applications. Note that the originator is generally described as engaging TGF-beta 1 and TGF-beta 2; this analog is characterized here around the TGFB1 target listed on the card. TGFB1 encodes transforming growth factor beta 1, a pleiotropic cytokine that regulates cell proliferation, differentiation, extracellular matrix production, immune homeostasis, and tissue repair. It is secreted as a latent complex in which the mature growth factor is non-covalently associated with its latency-associated peptide (LAP); activation (by integrins, proteases, or mechanical force) releases the active dimer. Active TGF-beta 1 signals through the type II (TGFBR2) and type I (TGFBR1/ALK5) serine/threonine kinase receptors, driving phosphorylation of SMAD2/SMAD3, which partner with SMAD4 to regulate transcription. In disease, excess TGF-beta 1 promotes fibrosis, immunosuppression within the tumor microenvironment, and resistance to checkpoint blockade, making it an actively pursued therapeutic target.
