Background
This is an unconjugated, non-therapeutic recombinant analog of onartuzumab, an antibody directed against human MET (hepatocyte growth factor receptor, c-Met). Onartuzumab is distinctive as a monovalent, one-armed antibody engineered to block ligand-driven receptor dimerisation without triggering agonist activity; the "Human Fab - G1 - kappa" descriptor reflects this monovalent, single antigen-binding format. Supplied for research use only, it is intended as a target-binding reagent for laboratory work, not for human or veterinary use. Typical applications include use as a positive control or benchmark in MET binding and neutralisation assays, receptor-signalling studies, epitope and competition work, ADCC or antibody-drug-conjugate development pipelines, and preclinical in-vitro and in-vivo model systems. Material is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options), in an unconjugated format suitable for downstream labelling, and available in bulk milligram-to-gram quantities to support screening, assay standardisation, and scale-up. It is a research analog built around the MET target, not the clinical drug product. MET (UniProt P08581), also called c-Met or hepatocyte growth factor receptor, is a single-pass transmembrane receptor tyrosine kinase. Its only known ligand is hepatocyte growth factor/scatter factor (HGF/SF). Ligand binding promotes receptor dimerisation and trans-autophosphorylation of the intracellular kinase domain, creating docking sites that recruit adaptors such as GRB2 and GAB1 and activate the RAS-MAPK, PI3K-AKT, and STAT3 pathways. This signalling drives cell proliferation, survival, motility, scattering, invasion, and branching morphogenesis, and is essential in embryonic development, liver regeneration, and wound healing. Aberrant MET activation, through gene amplification, activating mutations, protein overexpression, or autocrine/paracrine HGF loops, is implicated in tumour growth, invasion, metastasis, and resistance to other targeted therapies, making MET a widely studied oncology target.
