Background
This product is an unconjugated, non-therapeutic recombinant analog of imsidolimab, an anti-IL-36 receptor (IL-36R / IL1RL2) monoclonal antibody, produced for research use only. It reproduces the target specificity of the originator biologic (a humanized IgG4 antibody that binds IL-36R and antagonizes IL-36 signaling) in a research-grade format, and it is not the clinical drug and is not intended for human or veterinary use. The recombinant format makes it useful as a reference reagent and positive control in binding, blocking, and neutralization assays against human IL-36R, for benchmarking in-house or biosimilar candidate antibodies, and for supporting antibody-engineering and preclinical characterization work. It is supplied unconjugated so researchers can pair it with their own detection systems. Low-endotoxin grades (research grade under 1 EU/mg; ultra-low under 0.5 EU/mg) and bulk milligram-to-gram quantities are available to support cell-based assays, screening cascades, and scale-up. Because IL-36R is a human target, the reagent is oriented toward functional and in-vitro applications rather than routine rodent in-vivo dosing.
IL1RL2 (Q9HB29), also called IL-36R, IL-1Rrp2, or IL1RRP2, is a member of the interleukin-1 receptor family. It is the ligand-binding receptor for the IL-36 cytokines IL-36 alpha, IL-36 beta, and IL-36 gamma. On ligand engagement, IL-36R recruits the shared accessory protein IL1RAP (IL-1RAcP) to form a heterodimeric signaling complex that drives MyD88-dependent activation of NF-kappaB and MAPK pathways, producing proinflammatory responses. Signaling is normally restrained by the endogenous antagonist IL-36Ra, which binds IL-36R without recruiting the coreceptor. IL-36R is expressed prominently in epithelial and barrier tissues, particularly skin, where the IL-36 axis amplifies the IL-23/IL-17/IL-22 pathway. Dysregulated IL-36R signaling is central to pustular skin inflammation, including generalized pustular psoriasis and palmoplantar pustulosis, making the receptor a key node in epithelial autoinflammation.
