Background
This product is an unconjugated, non-therapeutic recombinant analog of datopotamab, supplied for research use only. Datopotamab is a humanised IgG1-kappa antibody directed against TACSTD2 (Trop-2), and in its clinical form it serves as the targeting component of an antibody-drug conjugate. This RUO analog reproduces the antigen-binding specificity for human Trop-2 in a naked (unconjugated) format, making it a useful tool for characterising Trop-2 biology and for benchmarking Trop-2-directed reagents. Typical applications include use as a binding and specificity control, as a positive reference in flow cytometry, ELISA, and immunoassays on Trop-2-expressing cells, and as a scaffold for in-house antibody-drug conjugate development and payload/linker chemistry. It also supports assay standardisation and comparison against biosimilar or competitor material. The recombinant human IgG1-kappa framework provides a defined, reproducible reagent. It is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options), and in bulk milligram-to-gram quantities suitable for in-vitro workflows and preclinical standards. It is not the clinical drug and is not for human or veterinary use.
TACSTD2 (tumour-associated calcium signal transducer 2), commonly called Trop-2 and also known as GA733-1 or EGP-1, is a single-pass transmembrane glycoprotein encoded by an intronless gene. It has a large cystine-rich extracellular domain, a single transmembrane region, and a short cytoplasmic tail containing a phosphorylation site and a PIP2-binding motif. Trop-2 participates in intracellular calcium signalling and transduces signals that influence cell proliferation, adhesion, and migration. Regulated intramembrane proteolysis can release an intracellular fragment that engages downstream pathways. Trop-2 is expressed at modest levels in many normal epithelia but is markedly overexpressed across numerous carcinomas, including breast (notably triple-negative), lung, and other epithelial tumours. This tumour-associated overexpression, combined with cell-surface accessibility, makes Trop-2 an attractive target for antibody and antibody-drug-conjugate approaches, and a frequent subject of expression and signalling studies.
