Ly-6G (lymphocyte antigen 6 complex, locus G; UniProt P35461) is a GPI-anchored cell-surface protein of the Ly-6 superfamily expressed on mouse myeloid cells. Its expression is tightly linked to granulocytic differentiation: it appears during neutrophil maturation in the bone marrow and is highly and selectively expressed on mature circulating and tissue neutrophils. Unlike the shared Gr-1 epitope (Ly-6C/Ly-6G) recognized by clones such as RB6-8C5, Ly-6G is not appreciably expressed on monocytes, making it a far more specific marker for the neutrophil lineage. Ly-6G is frequently used to identify granulocytic myeloid-derived suppressor cells (G-MDSC/PMN-MDSC) in tumor and inflammatory settings. While Ly-6G serves as a robust lineage marker, its precise endogenous signaling role is less well defined; it has been implicated in modulating neutrophil migration and function. Because expression is restricted to granulocytes, Ly-6G is a preferred target for selective neutrophil identification and depletion in vivo.
