Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-CD37 antibody naratuximab, produced for research use only. It is a full-length human IgG1/kappa immunoglobulin built around the CD37 target specificity of the originator, supplied naked (no maytansinoid payload or other conjugation) so that antigen binding can be studied independently of any cytotoxic warhead. Because naratuximab is best known as the backbone of an antibody-drug conjugate, an unconjugated analog is a useful tool for dissecting the contributions of target engagement versus payload delivery, and for serving as a matched building block in ADC-development and linker/payload optimization workflows. It is offered at research grade with low endotoxin (typically less than 1 EU/mg, with an ultra-low option under 0.5 EU/mg) and is available in bulk milligram-to-gram quantities suitable for in-vitro assays, binding and neutralization studies, isotype-matched controls, and preclinical model work. This is not the clinical drug and is not intended for human or veterinary use.
CD37 (UniProt P11049) is a heavily glycosylated four-transmembrane protein of the tetraspanin superfamily. Its expression is tightly restricted within the hematopoietic compartment, appearing on B cells from the pre-B through mature peripheral stages and lost upon terminal differentiation to plasma cells, with little to no expression on T cells, NK cells, monocytes, or granulocytes. Like other tetraspanins, CD37 organizes the plasma membrane into tetraspanin-enriched microdomains through lateral associations with partner tetraspanins and integrins. It is signaling-competent, carrying ITIM-like and ITAM-like intracellular motifs that couple to PI3K/AKT and related pathways to modulate both pro-survival and pro-apoptotic outputs. CD37 contributes to germinal center function, long-lived antibody production, B-cell survival, and immune regulation. Its B-lineage-restricted expression underlies its interest as a target in B-cell malignancies.
