Background
This product is an unconjugated, non-therapeutic recombinant analog of lilotomab, an anti-CD37 monoclonal antibody, supplied for research use only. It is a human IgG1/kappa antibody built to engage CD37 on the surface of normal and malignant B cells, and is not the clinical drug or a radioconjugate; it is intended as a naked (unlabelled) binder for laboratory work. The originator lilotomab is best known as the targeting antibody of the CD37-directed radioimmunoconjugate developed for B-cell non-Hodgkin lymphoma, where the cold antibody is also used as a pre-dose. In a research setting the unconjugated analog is useful as a defined anti-CD37 reagent for binding and specificity assays, flow-cytometry-based target detection, competition and epitope studies, as a positive or isotype-matched control, and as a starting antibody for antibody-drug-conjugate or radioimmunoconjugate development workflows. It is offered at research grade with low endotoxin (typically under 1 EU/mg, with ultra-low-endotoxin options under 0.5 EU/mg) and in bulk milligram-to-gram quantities suitable for assay development and preclinical characterization. Not for human or veterinary use. CD37 (UniProt P11049) is a member of the tetraspanin (transmembrane 4) superfamily, a heavily glycosylated cell-surface glycoprotein with four transmembrane domains and two extracellular loops. It is strongly and selectively expressed on mature B cells, with lower expression on other leukocytes, and is a structural component of tetraspanin-enriched microdomains that organize receptor clustering and signaling at the membrane. Through these interactions CD37 contributes to B-cell activation, adhesion and migration, survival/apoptotic signaling, and to germinal-center function and long-lived antibody responses; CD37-deficient mice show reduced IgG1 and impaired T-dependent antibody responses. CD37 is highly expressed on most B-cell non-Hodgkin lymphomas and its expression is independent of CD20-directed therapy, making it an attractive alternative antigen. It internalizes on antibody binding, supporting its use in conjugate-delivery strategies.
