Background
This product is a research-grade biosimilar of bermekimab (development codes MABp1; originator trade name Xilonix), supplied as an unconjugated, non-therapeutic analog built around the interleukin-1 alpha (IL-1A) target. It is offered for research use only and is not a clinical drug. Bermekimab is a fully human IgG1, kappa monoclonal antibody that binds the alarmin cytokine IL-1 alpha with high affinity and neutralises its biological activity, thereby preventing IL-1 alpha from engaging its receptor and driving sterile inflammation. The originator has been evaluated across advanced solid tumours (colorectal and pancreatic cancer), atopic dermatitis, and hidradenitis suppurativa, where IL-1 alpha-driven inflammation is implicated. This biosimilar reproduces the target specificity and human IgG1/kappa framework of the reference molecule so investigators can use it as a comparator, control, or tool reagent in functional assays. It is manufactured to research-grade endotoxin standards and available in bulk milligram-to-gram quantities. Interleukin-1 alpha (IL-1A; UniProt P01583) is a pleiotropic cytokine of the IL-1 family that bridges innate and adaptive immunity. It has a distinctive dual biology: constitutively present intracellularly, it can act in the nucleus, and upon cell damage or necrosis it is released as an alarmin that signals danger to the surrounding tissue, initiating sterile (non-infectious) inflammation. Secreted or membrane-associated IL-1 alpha binds the type I IL-1 receptor (IL1R1) together with the accessory protein IL1RAP, forming a high-affinity complex that activates NF-kB and the p38, ERK (p42/p44), and JNK MAPK pathways. This drives downstream pro-inflammatory gene expression. IL-1 alpha is implicated in tumour-associated inflammation, cardiovascular disease, and inflammatory skin conditions, making it an attractive therapeutic and research target for neutralising antibodies such as bermekimab.
