Background
This product is a research-grade biosimilar of sevirumab (also known as MSL-109), supplied as an unconjugated, non-therapeutic analog of the originator antibody for research use only. It is a fully human IgG1, kappa immunoglobulin built around the antigen recognized by the originator, the human cytomegalovirus (HCMV) envelope glycoprotein H (gH). It is not the licensed clinical drug and is not intended for diagnostic or therapeutic use. Sevirumab was originally developed as a neutralizing antibody for the prophylaxis and treatment of HCMV infection in immunocompromised populations, such as patients with AIDS-associated CMV retinitis and transplant recipients. This biosimilar reproduces the target-binding activity of that molecule and is intended as a reference standard and functional tool for in-vitro work. Typical applications include neutralization and binding assays against HCMV gH, use as an isotype-matched or benchmark control, and characterization of antibody-antigen interactions. Because the target is a viral human-pathogen glycoprotein rather than a mouse antigen, the reagent is characterized primarily by functional and in-vitro performance rather than by an in-vivo mouse dosing regimen.
Human cytomegalovirus (HCMV) glycoprotein H (gH) is an envelope glycoprotein that, together with glycoprotein L (gL) and additional accessory proteins, forms the gH/gL-based complexes central to viral entry. The gH/gL heterodimer serves as the core of both the trimeric complex (with gO) and the pentameric complex (with UL128/UL130/UL131A), which govern fusion and cell tropism across fibroblasts, epithelial, endothelial, and myeloid cells. gH is required for membrane fusion and is a major target of virus-neutralizing antibodies, making it an important focus for antiviral antibody development and vaccine design. Sevirumab binds gH-containing complexes and blocks HCMV entry and spread, neutralizing viral replication in vitro. HCMV is a widespread beta-herpesvirus that causes serious disease in immunocompromised individuals and is a leading infectious cause of congenital disease, underscoring the biological relevance of gH-directed reagents.
