Background
This product is an unconjugated, non-therapeutic recombinant analog of bapineuzumab, supplied strictly for research use only (RUO); it is not the clinical drug and is not intended for human or veterinary use. Bapineuzumab is a humanized IgG1 antibody directed against the amyloid-beta (Abeta) peptide, a proteolytic fragment derived from the amyloid precursor protein (APP, UniProt P05067). The research-grade analog reproduces the target-binding activity of the originator, making it useful as a defined reagent for in-vitro and preclinical work: positive and reference controls in Abeta binding and immunoassay development, benchmarking of anti-amyloid antibody panels, epitope-mapping studies, and as a comparator in neutralization or aggregation assays. It is manufactured for research workflows with low-endotoxin grades (research grade typically less than 1 EU/mg; ultra-low grade less than 0.5 EU/mg) and can be produced in bulk milligram-to-gram quantities to support assay standardization and larger preclinical studies. Being unconjugated, it also serves as a starting point for labeling, ADC-style conjugation development, or format-conversion research. Amyloid precursor protein (APP; UniProt P05067) is a large, ubiquitously expressed type I transmembrane glycoprotein with roles in synaptic formation, neurite outgrowth, cell adhesion, and neuronal signaling. APP is processed by two competing pathways: the non-amyloidogenic route (alpha-secretase then gamma-secretase) and the amyloidogenic route, in which beta-secretase (BACE1) followed by gamma-secretase liberates the amyloid-beta (Abeta) peptide (spanning roughly residues 672-713 of the APP770 isoform). Abeta, particularly the aggregation-prone Abeta42 species, accumulates into oligomers and extracellular plaques central to Alzheimer's disease pathology. Bapineuzumab recognizes the free N-terminus of the Abeta peptide (an epitope around residues 1-5), a neo-epitope exposed by APP cleavage rather than full-length APP broadly.
