Clone Cy34.1 is a mouse anti-mouse CD22 monoclonal antibody of the IgG1 isotype, raised against mouse CD22 (UniProt P35329). Because it recognises native cell-surface CD22 on B lineage cells, it is widely used for flow cytometric identification of B cells and, in functional studies, as a tool for interrogating the CD22 axis and B-cell targeting. This preparation is produced for in vivo and functional research: it is supplied at low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and can be provided in bulk milligram to gram quantities suitable for repeat-dose mouse studies and larger cohorts. As a mouse IgG1 reagent used in a mouse host, it is appropriate for studies where a syngeneic isotype is preferred and where Fc-mediated effector engagement is comparatively limited relative to IgG2a/IgG2b. CD22 (Siglec-2) is a B-cell-restricted type I transmembrane sialic-acid-binding immunoglobulin-like lectin. It is expressed on mature B cells and is one of the most B-lineage-specific surface markers, largely absent from plasma cells and non-B lineages, which makes it a useful marker and target. CD22 recognises alpha-2,6-linked sialic acid ligands and functions primarily as an inhibitory co-receptor that modulates B-cell receptor (BCR) signalling. Its cytoplasmic tail carries immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that, upon phosphorylation, recruit the phosphatase SHP-1 to dampen BCR-triggered calcium flux and activation, raising the threshold for B-cell responses and contributing to self-tolerance. CD22 also participates in regulating B-cell survival and homing. Because it is B-cell restricted and rapidly internalises upon antibody binding, CD22 is an established target for B-cell-directed depletion and antibody-drug conjugate strategies.
