Background
ICH5582 is a research-grade, unconjugated recombinant analog of gosuranemab, a humanized anti-tau antibody (human IgG4/kappa) built around the microtubule-associated protein tau (MAPT) target, specifically the extracellular N-terminal region of tau. It is supplied as a non-therapeutic reagent for research use only (RUO) and is not the clinical drug; it is not intended for human or veterinary use. The molecule reproduces the target-binding specificity of the originator, making it useful as a defined, lot-consistent tool for tau-directed experiments: positive and reference controls in binding and neutralisation assays, benchmarking of in-house anti-tau antibodies, epitope and pathway studies, and as a standard in preclinical in-vitro and cell-based tauopathy models. Available as an IgG4 isotype-matched reagent, it supports comparisons where Fc effector engagement should be minimal. It is offered at low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and in bulk milligram-to-gram quantities to support assay development, screening cascades, and scale-up work without lot-to-lot variability confounding results.
MAPT (P10636) encodes tau, a microtubule-associated protein highly enriched in neuronal axons. Through its C-terminal microtubule-binding repeats, tau promotes tubulin polymerisation and stabilises microtubules, regulating microtubule dynamics, axonal transport, and neuronal polarity, while its N-terminal projection domain interacts with the plasma membrane and other partners. Alternative splicing generates six CNS isoforms (3R and 4R). Tau activity is tightly controlled by phosphorylation; pathological hyperphosphorylation reduces microtubule affinity and promotes detachment and self-aggregation into paired helical filaments and neurofibrillary tangles. These aggregates, together with cell-to-cell spread of misfolded and extracellular tau species, define the tauopathies, including Alzheimer's disease, progressive supranuclear palsy, and frontotemporal dementia. The N-terminal region targeted by gosuranemab corresponds to an extracellular tau species implicated in trans-neuronal propagation of tau pathology.
