Background
This is an unconjugated, non-therapeutic recombinant research analog of ongericimab, an anti-PCSK9 monoclonal antibody, supplied strictly for research use only. It is built around the well-established PCSK9 target and reproduces the binding specificity of the originator antibody, but it is not the clinical drug and is not intended for human or veterinary use. The analog is useful as a reference reagent for characterizing PCSK9 biology and antibody behavior in vitro: positive/isotype-matched binding controls, epitope-competition and cross-blocking assays, surface-plasmon-resonance or biolayer-interferometry affinity work, ELISA and flow standards, and as a benchmarking tool when developing or screening other PCSK9-directed binders. It also supports preclinical mechanism-of-action studies of the PCSK9-LDLR axis in cell-based systems. Material is offered as low-endotoxin, bulk research grade (research grade <1 EU/mg; ultra-low <0.5 EU/mg) with milligram-to-gram scale availability suited to assay development and in-vitro standardization. No clone identity is implied. Users should validate performance in their own systems before relying on it as a quantitative standard.
PCSK9 (proprotein convertase subtilisin/kexin type 9; UniProt Q8NBP7) is a secreted serine protease of the proteinase K subfamily that is a master regulator of circulating LDL cholesterol. It is expressed mainly in liver, intestine, and kidney. After autocatalytic cleavage of its prodomain in the endoplasmic reticulum, the prodomain remains non-covalently associated and PCSK9 is secreted as a mature, catalytically inactive chaperone. Circulating PCSK9 binds the EGF-A domain of the hepatic LDL receptor (LDLR) at the cell surface; this interaction redirects the internalized LDLR to lysosomal degradation rather than recycling, lowering surface LDLR density and raising plasma LDL-C. Gain-of-function PCSK9 variants cause autosomal dominant hypercholesterolemia, whereas loss-of-function variants lower LDL-C and cardiovascular risk. Antibodies that block the PCSK9-LDLR interaction spare LDLR from degradation, increasing LDL clearance.
