Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-PCSK9 antibody ebronucimab, supplied for research use only. Ebronucimab is a fully human IgG1 antibody (originator Akeso) that binds proprotein convertase subtilisin/kexin type 9 (PCSK9) and blocks its interaction with the LDL receptor. The research-grade analog is built around the same human PCSK9 target and reproduces that binding activity, but it is not the clinical drug and is not intended for human or veterinary use. It is useful as an isotype-matched (human IgG1/lambda) tool for PCSK9-directed assays: a positive binding and neutralisation control, a benchmark or comparator in PCSK9-inhibitor screening campaigns, a reference reagent for developing and validating PCSK9/LDLR interaction, ELISA, SPR/BLI and cell-based LDL-uptake assays, and a starting reagent for ADC or engineering workflows. It is offered at research grade with low endotoxin (typically less than 1 EU/mg, with ultra-low-endotoxin below 0.5 EU/mg options) and is available in bulk milligram-to-gram quantities for in-vitro and preclinical work. PCSK9 (proprotein convertase subtilisin/kexin type 9; UniProt Q8NBP7) is a secreted serine protease produced mainly by hepatocytes. After autocatalytic cleavage in the ER, it circulates as a mature enzyme whose prodomain remains non-covalently associated with the catalytic domain. Rather than acting through proteolysis on the LDL receptor (LDLR), PCSK9 functions largely as a chaperone: it binds the EGF-A domain of surface LDLR, and after receptor-mediated endocytosis it prevents the pH-dependent conformational change that normally releases LDL, redirecting the LDLR-PCSK9 complex to lysosomal degradation instead of recycling. The net effect is fewer surface LDL receptors, reduced hepatic LDL clearance, and higher plasma LDL cholesterol. Gain-of-function PCSK9 variants cause familial hypercholesterolemia, whereas loss-of-function variants lower LDL-C and cardiovascular risk, making PCSK9 a validated lipid-lowering target.
