Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-mesothelin antibody misitatug (the antibody component of the mesothelin-directed antibody-drug conjugate program, e.g. RC88 / misitatug blivedotin). It is built around the human mesothelin (MSLN) target and supplied as a research-grade human IgG1-kappa protein for research use only. It is not the clinical drug and is not intended for human or veterinary use. The molecule is unconjugated (no payload, no label), which makes it useful as a naked-antibody control, as a benchmarking reagent against other anti-MSLN binders, and as a defined starting material for ADC and conjugation development workflows. Typical research applications include target-binding and specificity studies on MSLN-expressing cell lines, neutralization and blocking assays, epitope and affinity characterization, flow cytometry, and use as a positive/isotype-matched reference in assay development. It is offered at low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and available in bulk milligram-to-gram quantities to support in-vitro and preclinical study scale-up. Mesothelin (MSLN, UniProt Q13421) is a glycosylphosphatidylinositol (GPI)-anchored cell-surface glycoprotein. It is synthesized as a ~71 kDa precursor that is proteolytically processed by furin into a secreted ~31 kDa N-terminal fragment, megakaryocyte potentiating factor (MPF), and a membrane-bound ~40 kDa mature mesothelin. Mature mesothelin binds the mucin MUC16 (CA125), an interaction implicated in cell adhesion and in the peritoneal spread of tumor cells. In normal tissue, mesothelin expression is largely restricted to mesothelial cells lining the pleura, peritoneum and pericardium. It is strongly overexpressed in several solid tumors, including malignant mesothelioma, pancreatic, ovarian, lung and biliary tract cancers. This tumor-associated, cell-surface localization, with limited normal-tissue expression, is the rationale for mesothelin as a target for antibodies, antibody-drug conjugates, immunotoxins and CAR-T approaches.
