The T cell receptor (TCR) is a cell-surface antigen-recognition complex expressed on T lymphocytes. In most circulating T cells it consists of a disulfide-linked alpha and beta chain heterodimer, each with a variable and a constant domain, generated through V(D)J recombination to produce a highly diverse repertoire. The TCR recognises short peptide antigens presented by major histocompatibility complex (MHC) molecules on other cells. Because the alpha/beta heterodimer itself has short cytoplasmic tails and cannot signal alone, it associates non-covalently with the invariant CD3 chains (gamma, delta, epsilon) and the zeta homodimer, which carry the ITAM motifs that transduce activation signals. Engagement of the TCR-CD3 complex drives T cell activation, proliferation, and differentiation, making it central to adaptive immunity and a target of interest for immunomodulation and immuno-oncology research.
