Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-ROR2 antibody ozuriftamab, produced for research use only. It reproduces the antigen-binding specificity of the originator molecule directed against human ROR2 (UniProt Q01974) on a human IgG1-kappa framework, but is supplied naked (no cytotoxic payload or conjugate) and is not intended for any human or veterinary therapeutic application. The originator antibody is being developed in an antibody-drug-conjugate format targeting ROR2, a tumor-associated receptor tyrosine kinase; this analog is useful for dissecting the biology and binding behavior of that target without the confounding effects of a payload. As a bulk, well-characterized reagent it supports antigen-binding and epitope studies, use as a positive/isotype-matched control, flow cytometry and immunoassay development, and as a targeting-antibody scaffold for ADC and effector-function development workflows. It is manufactured at low endotoxin (research grade <1 EU/mg, ultra-low <0.5 EU/mg) and available in preclinical in-vitro standard and bulk milligram-to-gram quantities to support reproducible, scalable experiments. RUO; not the clinical drug. ROR2 (receptor tyrosine kinase-like orphan receptor 2; UniProt Q01974) is a single-pass transmembrane receptor of the ROR family, containing an extracellular immunoglobulin-like domain, a cysteine-rich (Frizzled-like) domain, and a Kringle domain, plus an intracellular tyrosine kinase domain that is catalytically atypical. It is the principal receptor for the non-canonical Wnt ligand Wnt5a, and together with Frizzled co-receptors it transduces beta-catenin-independent signaling through the planar cell polarity and Wnt/Ca2+ pathways, regulating cell polarity, directional migration, and cytoskeletal organization. ROR2 is highly expressed during embryonic skeletal and cardiac development and is largely silenced in most normal adult tissues. Its re-expression is associated with invasion and metastasis in several tumor types, including melanoma, renal cell, ovarian, and gastrointestinal cancers, making it an attractive tumor-associated antigen.
