It is a fully human IgG1-kappa antibody built around Delta-like ligand 4 (DLL4), the Notch pathway ligand targeted by the originator molecule. Delta-like ligand 4 (DLL4; UniProt Q9NR61) is a single-pass transmembrane ligand of the Notch signaling pathway, expressed predominantly by arterial and tip endothelial cells. Binding of DLL4 to Notch1/Notch4 receptors on neighboring cells triggers receptor cleavage and release of the Notch intracellular domain, which drives transcription of downstream genes. In the vasculature, DLL4-Notch signaling is a central regulator of sprouting angiogenesis: it restrains excessive tip-cell formation and vessel branching, thereby promoting the formation of functional, well-perfused vasculature. Paradoxically, blocking DLL4 increases vessel sprouting but yields non-productive, poorly perfused networks, an effect exploited in oncology research. DLL4 also contributes to arterial specification and to immune and stem-cell regulation. Its dysregulation is implicated in tumor angiogenesis, making it an actively studied target.
