Background
This product is an unconjugated, non-therapeutic recombinant analog of enoticumab, supplied for research use only. It is a fully human IgG1-kappa antibody built around Delta-like ligand 4 (DLL4), the Notch pathway ligand targeted by the originator molecule. It is not the clinical drug and is not intended for human or veterinary use. The analog reproduces the binding specificity of the parent antibody, making it a practical tool for laboratory work: as a positive control in DLL4 binding and blocking assays, as a reference reagent when benchmarking novel anti-DLL4 antibodies or bispecifics, as a starting point for antibody-drug conjugate or ADCC/CDC assay development, and for target-engagement studies in cell-based systems. Supplied unconjugated so users can label or reformat as needed. Available at research-grade low endotoxin (<1 EU/mg) and ultra-low endotoxin (<0.5 EU/mg) grades, and in bulk milligram-to-gram quantities for larger screening campaigns and preclinical model work. Because it is a research analog rather than the formulated therapeutic, buffer, concentration and endotoxin can be matched to assay requirements. Delta-like ligand 4 (DLL4; UniProt Q9NR61) is a single-pass transmembrane ligand of the Notch signaling pathway, expressed predominantly by arterial and tip endothelial cells. Binding of DLL4 to Notch1/Notch4 receptors on neighboring cells triggers receptor cleavage and release of the Notch intracellular domain, which drives transcription of downstream genes. In the vasculature, DLL4-Notch signaling is a central regulator of sprouting angiogenesis: it restrains excessive tip-cell formation and vessel branching, thereby promoting the formation of functional, well-perfused vasculature. Paradoxically, blocking DLL4 increases vessel sprouting but yields non-productive, poorly perfused networks, an effect exploited in oncology research. DLL4 also contributes to arterial specification and to immune and stem-cell regulation. Its dysregulation is implicated in tumor angiogenesis, making it an actively studied target.
