Background
Anrukinzumab Biosimilar (Research Grade) is an unconjugated, non-therapeutic research analog of anrukinzumab (also known as IMA-638), a humanized IgG1 kappa antibody directed against human interleukin-13 (IL-13). It is manufactured for research use only and is not a clinical drug. The originator anrukinzumab was developed to neutralize soluble IL-13, a central type 2 (Th2) cytokine, and was investigated clinically in allergic asthma and ulcerative colitis. Mechanistically, the antibody binds IL-13 and blocks its ability to recruit IL-4Ralpha into a signaling complex with IL-13Ralpha1, thereby preventing downstream STAT6 activation without interfering with IL-13 binding to IL-13Ralpha1 or the decoy receptor IL-13Ralpha2. As a research-grade biosimilar, this product is intended as a functional tool and comparator reagent for in-vitro characterization of IL-13 biology, cytokine-neutralization assays, and biosimilarity or bioanalytical method development. It is supplied at low endotoxin levels and is available in bulk milligram to gram quantities to support reproducible, well-controlled experimental work built around the IL-13 target. Interleukin-13 (IL-13, UniProt P35225) is a ~146-amino-acid secreted type 2 cytokine produced largely by activated Th2 cells, as well as mast cells, basophils, eosinophils and ILC2s. It shares many biological activities with IL-4 because both signal through the type II receptor complex. IL-13 binds IL-13Ralpha1, which then recruits IL-4Ralpha to trigger JAK/STAT6 signaling, driving IgE class switching, goblet-cell mucus production, airway smooth-muscle responses, fibrosis and epithelial changes. A second receptor, IL-13Ralpha2, acts as a high-affinity decoy that limits signaling. IL-13 is a major driver of allergic and type 2 inflammatory disease, making it a validated therapeutic and research target in asthma, atopic dermatitis and fibrotic and gastrointestinal inflammatory conditions.
