Background
The Amlitelimab Biosimilar - Research Grade (ICH5226) is an unconjugated, non-therapeutic recombinant analog of the originator anti-OX40L antibody amlitelimab, built as a fully human IgG4 kappa immunoglobulin directed against human TNFSF4 (OX40 ligand, CD252). It is supplied for research use only and is not the clinical drug, not for human or veterinary use. The molecule is intended as a reference binder and functional tool for investigators studying the OX40-OX40L costimulatory axis, allowing researchers to probe ligand blockade, receptor-ligand interaction, and T-cell costimulation in defined systems. Typical applications include use as a positive/isotype-matched control, a capture or detection reagent in binding assays, a neutralizing antibody in cell-based costimulation readouts, and a benchmarking standard for assay development and comparability work. Because it mirrors an IgG4 backbone, it is useful for studying Fc-silent, non-depleting engagement of a TNF-superfamily ligand. It is offered at low endotoxin (research grade less than 1 EU/mg; ultra-low less than 0.5 EU/mg) and in bulk milligram-to-gram quantities to support in-vitro screening and preclinical workflows. TNFSF4 (OX40 ligand, OX40L, CD252, gp34; UniProt P23510) is a type II transmembrane member of the TNF ligand superfamily that assembles as a homotrimer and is expressed on activated antigen-presenting cells such as dendritic cells, B cells, and macrophages, as well as on activated endothelial and mast cells. Its receptor, OX40 (TNFRSF4, CD134), is induced on activated CD4+ and CD8+ T cells. Engagement of OX40 by OX40L delivers a late costimulatory signal that promotes T-cell proliferation, survival, cytokine production, and effector/memory differentiation, and it influences regulatory T-cell function. The pathway amplifies type 2 as well as Th1, Th17, and Th22 responses, and OX40L can also transmit reverse (back) signaling into ligand-bearing cells. Genetic variation at the TNFSF4 locus has been linked to autoimmune conditions including systemic lupus erythematosus and Sjogren syndrome, making the axis a focus of inflammation and autoimmunity research.
