Background
This product is an unconjugated, non-therapeutic recombinant analog of zagotenemab, supplied for research use only. It is a full-length recombinant human IgG4 (kappa) antibody engineered to reproduce the antigen-binding specificity of the originator anti-tau molecule, which recognizes a pathological, misfolded conformation of the microtubule-associated protein tau associated with Alzheimer's disease and related tauopathies. It is not the clinical drug and is not intended for human or veterinary use. The IgG4 backbone carries stabilizing and effector-attenuating substitutions (S228P to prevent Fab-arm exchange; F234A/L235A to reduce Fcgamma-receptor engagement) plus a C-terminal lysine deletion (K447del) for charge homogeneity, making it a clean reagent for conformation-selective binding studies. Typical research applications include use as a binding and neutralization reference standard, an isotype-matched control in tau immunoassays, an antigen-capture or detection reagent, and a tool for characterizing aggregated versus monomeric tau species. It is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg option), and available in bulk milligram-to-gram quantities for assay development and preclinical work. Tau (MAPT gene product, UniProt P10636) is a microtubule-associated protein enriched in neuronal axons, where it binds and stabilizes microtubules and regulates axonal transport. Human tau exists as six main splice isoforms differing in the number of N-terminal inserts and microtubule-binding repeats (3R versus 4R). Its microtubule affinity is dynamically controlled by phosphorylation across numerous serine/threonine sites. In Alzheimer's disease and other tauopathies, tau becomes hyperphosphorylated and adopts pathological, misfolded conformations, detaching from microtubules and self-aggregating into paired helical filaments and neurofibrillary tangles. Misfolded tau can propagate between neurons in a prion-like manner, seeding further aggregation. Conformation-selective antibodies aim to distinguish these pathological species from normal, functional tau, making tau a central target for both diagnostic reagents and disease-modifying antibody research.
