Background
This is a research-grade, unconjugated recombinant analog of naxitamab, an anti-GD2 antibody, supplied for research use only and not for human or veterinary use. It reproduces a naxitamab-style human IgG1 Kappa format (carrying E356D/M358L allotype substitutions) that binds the disialoganglioside GD2, giving labs a non-therapeutic tool to study GD2 engagement without sourcing the clinical drug. Typical applications include a positive binding control for GD2-expressing cell lines, a benchmark reagent in ELISA/flow/SPR assays, a reference for Fc-effector studies such as ADCC and CDC, and a starting point for antibody-drug-conjugate or CAR development against GD2. It is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options), as an in-vivo-standard where applicable, and in bulk milligram-to-gram quantities to support screening, assay development, and preclinical work. Because it is built around the target rather than being the formulated therapeutic, it should be treated as a characterization and control reagent, not a clinical product. GD2 is a disialoganglioside, a glycosphingolipid bearing two sialic acid residues, embedded in the outer leaflet of the plasma membrane. In healthy tissue its expression is largely restricted to neurons, peripheral nerves, and skin melanocytes, but it is strongly and uniformly overexpressed on neuroblastoma and on many melanomas, retinoblastomas, small-cell lung cancers, and certain sarcomas. This tumor-associated, near-lineage-restricted distribution makes GD2 an attractive immunotherapy target. As a lipid antigen it lacks the mobility and shedding behavior of many protein targets and is not internalized efficiently, so anti-GD2 antibodies act largely through Fc-mediated effector functions (ADCC and complement-dependent cytotoxicity) rather than signaling blockade. GD2 also contributes to tumor cell adhesion, proliferation, and invasion. Its restricted expression underlies both the therapeutic rationale and the on-target neuropathic pain seen clinically with anti-GD2 agents.
