Background
This product is an unconjugated, non-therapeutic recombinant analog of lirilumab, supplied for research use only. Lirilumab is a fully human IgG4 antibody directed against inhibitory killer-cell immunoglobulin-like receptors of the KIR2DL family (KIR2DL1, and cross-reactively KIR2DL2/KIR2DL3), with reported cross-recognition of the activating KIR2DS1/KIR2DS2 receptors; it blocks the KIR-HLA-C interaction to lower the activation threshold of NK cells. This analog reproduces the antigen-binding specificity of the originator against the KIR2D target and is built around that target rather than being the clinical drug. The IgG4 backbone carries the S228P hinge-stabilizing mutation to limit half-antibody exchange. It is useful as an isotype-matched binding and blocking reagent, a reference standard for NK-cell checkpoint assays, a positive control in flow cytometry and receptor-occupancy work, and a tool for in-vitro and preclinical mechanistic studies. Offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options) and in bulk milligram-to-gram quantities. Not for human or veterinary use. KIR2DL1 is an inhibitory killer-cell immunoglobulin-like receptor expressed on natural killer (NK) cells and subsets of T cells. It belongs to the KIR family of type I transmembrane glycoproteins that regulate NK-cell effector function through recognition of HLA class I molecules. KIR2DL1 has two extracellular Ig-like domains and recognizes HLA-C allotypes bearing a lysine at position 80 (group C2). Ligand engagement triggers phosphorylation of cytoplasmic immunoreceptor tyrosine-based inhibitory motifs (ITIMs), which recruit SHP-1/SHP-2 phosphatases and deliver an inhibitory signal that dampens NK-cell cytotoxicity and cytokine release. This "missing-self" surveillance spares healthy HLA-C-expressing cells while licensing NK responses against cells that downregulate class I, such as some tumor and virally infected cells. The closely related KIR2DL2 and KIR2DL3 recognize the C1 group, and blocking these inhibitory receptors is a strategy to unleash NK-mediated antitumor activity.
