Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-NGF antibody fasinumab, engineered around human nerve growth factor (NGF, UniProt P01138) as its target. It is supplied for research use only and is not the clinical drug, nor intended for human or veterinary use. Built on a human IgG4 (S228P-stabilized) kappa framework, it reproduces the antigen-binding specificity of the originator against NGF, making it a convenient tool for interrogating NGF-driven signaling and for benchmarking assay performance. Typical research applications include use as a neutralizing reagent, a positive/reference control in binding and blocking assays, a capture or detection reagent in ligand-based ELISAs, and a comparator in biosimilarity or developability workflows. It is offered in low-endotoxin research grade (<1 EU/mg) and ultra-low-endotoxin (<0.5 EU/mg) formats, and is available in bulk milligram-to-gram quantities to support in-vitro screening, cell-based assays, and preclinical model work where large, consistent lots are needed. Storage and handling follow standard antibody practice; buffer and formulation details are lot-specific. Nerve growth factor (NGF) is the prototypical neurotrophin, secreted as a precursor (proNGF) and processed to mature beta-NGF, which functions as a non-covalent homodimer. It signals through two receptors: the high-affinity receptor tyrosine kinase TrkA (NTRK1), driving neuronal survival, differentiation, and axonal growth via Ras/MAPK, PI3K/Akt, and PLC-gamma pathways; and the low-affinity p75 neurotrophin receptor (p75NTR/NGFR), which modulates affinity and can trigger apoptotic or survival signals depending on context. Beyond its classical role in sympathetic and sensory neuron development, NGF is a key mediator of peripheral pain sensitization and inflammatory hyperalgesia, acting on nociceptors and immune cells. Elevated NGF is implicated in chronic pain conditions such as osteoarthritis and chronic low back pain, which is why NGF neutralization has been pursued therapeutically. NGF is highly conserved across mammals.
