Background
ICH5615 is a research-grade, unconjugated recombinant analog of the anti-NGF antibody izenivetmab, built around human nerve growth factor (beta-NGF, UniProt P01138). It is a non-therapeutic biosimilar intended strictly for research use, not for human or veterinary use, and is not the clinical drug itself. Expressed as a human IgG2-kappa immunoglobulin, it reproduces the target specificity of the originator so investigators can study NGF neutralisation, NGF/receptor interactions, and antibody-based blockade in controlled experimental systems. Typical research applications include use as a well-defined positive control or reference antibody, as a neutralising reagent in cell-based and biochemical assays, and as a tool for developing binding, ELISA, and ligand-blocking readouts. The material is supplied unconjugated, allowing downstream labelling or format conversion by the end user. Nerve growth factor (NGF; beta-NGF, UniProt P01138) is the prototypic neurotrophin, secreted as a mature ~120 aa protein that signals as a non-covalent homodimer. It binds two receptors: the high-affinity receptor tyrosine kinase TrkA (NTRK1), which drives neuronal survival, differentiation, and neurite outgrowth via Ras-MAPK, PI3K-Akt, and PLCgamma pathways, and the low-affinity p75 neurotrophin receptor (NGFR/TNFRSF16), which modulates affinity and can influence apoptotic and survival signalling. NGF is essential for the development and maintenance of sympathetic and sensory neurons. In adults it is a key mediator of nociceptor sensitisation and pain: NGF released during inflammation and tissue injury sensitises peripheral sensory neurons, upregulates pain-related channels and neuropeptides, and contributes to chronic and inflammatory pain states. This nociceptive role underlies the therapeutic interest in NGF-neutralising antibodies.
