Background
This product is an unconjugated, non-therapeutic recombinant analog of evinacumab, a fully human anti-ANGPTL3 antibody, supplied for research use only and not for human or veterinary use. It is a human IgG4 (S228P hinge-stabilized) kappa molecule built to recognize human angiopoietin-like 3 (ANGPTL3, UniProt Q9Y5C1), mirroring the target specificity of the originator drug without being the clinical product. It is useful as a defined, reproducible reagent for interrogating ANGPTL3 biology: binding and affinity characterization, neutralization/blockade assays against recombinant or native ANGPTL3, epitope and cross-reactivity studies, and as a positive control or benchmark antibody in assay development. The S228P substitution stabilizes the IgG4 hinge and limits Fab-arm exchange, giving consistent bivalent binding for structure-function and comparability work. Offered at research grade with low endotoxin (typically <1 EU/mg, with ultra-low <0.5 EU/mg options) and available in bulk milligram-to-gram quantities to support in-vitro screening, biochemical assays, and preclinical model work requiring lot-to-lot consistency. ANGPTL3 (angiopoietin-like 3) is a secreted, liver-derived glycoprotein and a central regulator of plasma lipid metabolism. It is a member of the angiopoietin-like family, comprising an N-terminal coiled-coil domain and a C-terminal fibrinogen-like domain. ANGPTL3 inhibits two key lipolytic enzymes: lipoprotein lipase (LPL), reducing hydrolysis of triglyceride-rich lipoproteins, and endothelial lipase (EL), which acts on HDL phospholipids. By restraining LPL and EL, ANGPTL3 raises circulating triglycerides, LDL cholesterol, and HDL cholesterol. It often acts in concert with ANGPTL8 to modulate LPL activity in a tissue- and feeding-state-dependent manner. Loss-of-function ANGPTL3 variants in humans produce familial combined hypolipidemia, with broadly reduced atherogenic lipids and apparent cardiovascular protection, which established ANGPTL3 as an attractive lipid-lowering target that acts independently of the LDL-receptor pathway.
