Background
Lesofavumab Biosimilar - Research Grade (ICH5637) is an unconjugated, non-therapeutic research analog of lesofavumab, a human anti-influenza B virus antibody originally developed by Genentech/Roche (also known as MHAB5553A or RG6024) and investigated for the treatment and prevention of influenza B infection. This product is built around the same target, influenza B virus hemagglutinin (HA), and is supplied as a human IgG1, kappa immunoglobulin for research use only. It is not a clinical drug and is not intended for diagnostic or therapeutic use. As a research-grade reagent it is manufactured to a low endotoxin specification (<1 EU/mg research grade; ultra-low <0.5 EU/mg options), which supports demanding cell-based and functional assays. Being an analog of a clinically studied antibody, it serves as a well-defined reference tool for characterizing anti-HA binding, neutralization, and Fc-mediated activity against influenza B, and for benchmarking novel antibodies, vaccine antigens, and assay platforms. Bulk quantities from milligram to gram scale can be supplied to support method development, standardization, and larger studies. The reagent is intended as a consistent, high-purity comparator rather than a substitute for the originator biologic. Influenza B virus hemagglutinin (HA) is the major surface glycoprotein that mediates viral attachment and entry. It is synthesized as an HA0 precursor that is cleaved into HA1 and HA2 subunits, assembling into a homotrimer with a membrane-distal globular head and a membrane-proximal stalk (stem). The head carries the sialic acid receptor-binding site and the most variable, immunodominant antigenic sites, so head-directed antibodies typically block receptor binding. The stalk is more conserved and drives pH-triggered membrane fusion in the endosome; stalk-directed antibodies tend to block fusion and can act through Fc-mediated effector functions such as ADCC. HA is the principal target of neutralizing antibodies against influenza B and a central antigen in vaccine and therapeutic antibody development, making it a key focus for neutralization and protection studies.
