CD52 (encoded by the CD52 gene; UniProt P31358) is a very small glycoprotein consisting of a short 12-amino-acid mature peptide anchored to the cell surface by a glycosylphosphatidylinositol (GPI) linkage and decorated with a large N-linked glycan. It is expressed abundantly on the surface of most mature lymphocytes, including T cells, B cells, natural killer cells, monocytes, macrophages, dendritic cells, and cells of the male reproductive tract, but is notably absent from hematopoietic stem cells. Despite its small size, CD52 is displayed at high copy number, making it an efficient target for antibody-mediated lysis via complement-dependent cytotoxicity and antibody-dependent cell-mediated cytotoxicity. Its precise physiological function is incompletely defined, though roles in T-cell costimulation, immune regulation, and modulation of complement have been proposed. Its broad lymphocyte expression underlies interest in CD52 as a target for profound, durable lymphocyte depletion.
