Background
This product is an unconjugated, non-therapeutic recombinant analog of enfortumab, supplied for research use only. It is a fully human IgG1 kappa antibody directed against human Nectin-4, produced recombinantly and provided in an unconjugated format (no drug payload attached), which distinguishes it from the clinical antibody-drug conjugate that inspired it. It is not the clinical drug and is not intended for human or veterinary use. The analog is useful as a target-binding reagent for studying Nectin-4 expression and function, as a positive or isotype-matched control in binding and blocking experiments, and as a starting antibody for antibody-drug conjugate (ADC) development, internalization studies, and characterization of anti-Nectin-4 activity. Because it reproduces the binding specificity of the originator against a well-characterized tumor-associated antigen, it supports flow cytometry, ELISA, immunoprecipitation, cell-based binding, and neutralization workflows. It is offered at research grade with low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and in bulk milligram-to-gram quantities suitable for in-vitro assays and preclinical models. Nectin-4 (poliovirus receptor-related 4, PVRL4; UniProt Q96NY8) is a type I transmembrane glycoprotein of the nectin family of immunoglobulin-like cell adhesion molecules. Its ectodomain contains three Ig-like domains that mediate calcium-independent cell-cell adhesion through homophilic and heterophilic interactions, notably trans-interaction with Nectin-1, contributing to adherens junction organization and epithelial integrity. Nectin-4 is highly expressed during embryonic development but shows restricted expression in most healthy adult tissues. It is markedly overexpressed in numerous carcinomas, including urothelial (bladder), breast, lung, and pancreatic cancers, where it correlates with proliferation, adhesion, and metastatic behavior and can be shed as a soluble form. This tumor-associated, developmentally regulated expression pattern makes Nectin-4 an attractive antigen for targeted therapy and a well-defined receptor for studies of tumor cell adhesion, signaling, and antibody-mediated targeting.
