Background
This product is an unconjugated, non-therapeutic recombinant analog of teprotumumab, supplied for research use only. It is not the clinical drug and is not intended for human or veterinary use. Teprotumumab is a fully human monoclonal antibody directed against the insulin-like growth factor 1 receptor (IGF1R); this analog reproduces that antigen-binding specificity so investigators can study IGF1R engagement and blockade in a controlled, reproducible reagent. The isotype is human IgG1 with kappa light chain, carrying E356D/M358L allotype substitutions in the Fc, which are relevant when matching allotype for Fc-dependent readouts. Typical research applications include IGF1R binding characterization, receptor-blockade and neutralization assays, positive and reference controls, and preclinical in-vitro and cell-based models. It is available bulk (mg to gram scale) and as low-endotoxin preparations (research grade <1 EU/mg; ultra-low <0.5 EU/mg) suitable for endotoxin-sensitive systems. Because the analog is defined by target specificity and format rather than a specific production clone, it serves as a consistent tool antibody for method development, assay standardization, and comparability work across experiments.
IGF1R (insulin-like growth factor 1 receptor; UniProt P08069) is a transmembrane receptor tyrosine kinase and the principal receptor for the growth factors IGF-1 and IGF-2. It is a disulfide-linked (alpha-beta)2 heterotetramer: the extracellular alpha subunits bind ligand, while the beta subunits contain the intracellular tyrosine kinase domain. Ligand binding triggers autophosphorylation and recruitment of IRS-1/IRS-2 and Shc, activating the PI3K-AKT and RAS-MAPK pathways to drive cell proliferation, growth, survival, and metabolism. IGF1R is closely related to the insulin receptor (INSR; UniProt P06213), with which it shares high homology and can form hybrid receptors, an important consideration for specificity. Dysregulated IGF1R signaling is implicated in oncology and, notably, in thyroid eye disease, where IGF1R functionally couples with the TSH receptor on orbital fibroblasts, the rationale underlying teprotumumab.
