Background
This product is an unconjugated, non-therapeutic recombinant analog of ozanezumab, an anti-Nogo-A (RTN4) antibody, supplied strictly for research use only. It is built around the same target and human IgG1-kappa framework concept as the originator biologic but is manufactured as a research reagent, not the clinical drug, and is not intended for human or veterinary use. The molecule is engineered with Fc-attenuating mutations, consistent with the originator's effector-reduced design, making it useful when researchers want target engagement without strong Fc-mediated effector function. Typical applications include use as a well-characterized positive or benchmarking control in binding and neutralization assays, as a tool for probing Nogo-A biology in vitro, in ADC or antibody-engineering workflows, and in preclinical model development. It is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options), and can be supplied in bulk milligram-to-gram quantities and as an in-vivo-standard grade to support scaled preclinical and assay-development programs. No clone identity is implied; the analog is defined by its target specificity and sequence framework.
Nogo-A is the largest isoform of Reticulon 4 (RTN4; UniProt Q9NQC3), a CNS-enriched membrane protein and one of the principal myelin-associated inhibitors of axonal growth and regeneration. Two functional regions drive its inhibitory activity: the amino-terminal Nogo-A-specific domain (including the Nogo-A-Delta20 region) and the Nogo-66 loop. Nogo-66 signals through Nogo receptor 1 (NgR1) with co-receptors such as p75/TROY and LINGO-1, while Nogo-A-Delta20 acts via sphingosine-1-phosphate receptor 2 (S1PR2). Engagement of these pathways activates RhoA/ROCK signaling, causing growth-cone collapse and restricting neurite outgrowth, plasticity, and circuit remodeling. Through these mechanisms Nogo-A contributes to the limited regenerative capacity of the adult mammalian CNS. It is studied extensively in spinal cord injury, stroke, and motor neuron disease, where elevated Nogo-A has been reported. RTN4 isoforms also localize to the endoplasmic reticulum and help shape ER membrane morphology.
