Background
This product is an unconjugated, non-therapeutic recombinant analog of gemtuzumab, an anti-CD33 antibody, supplied strictly for research use only and not for human or veterinary application. It is built around the CD33 (Siglec-3) target using a human IgG4 (S228P) kappa framework, matching the isotype of the originator so that Fc-dependent behavior is representative in experimental systems. The material is provided as a purified, carrier-free preparation suitable for laboratory workflows, with low-endotoxin research grade (<1 EU/mg) and ultra-low-endotoxin (<0.5 EU/mg) options, and it is available in bulk milligram-to-gram quantities to support scaled or repeated studies. Because it is a naked (unconjugated) analog, it is useful as a positive binding control, an isotype-matched reagent, a benchmarking tool against other anti-CD33 binders, and a starting scaffold for antibody-drug-conjugate (ADC) development or comparability work. CD33 (Siglec-3; UniProt P20138) is a transmembrane sialic-acid-binding immunoglobulin-type lectin expressed predominantly on cells of the myeloid lineage, including myeloid progenitors, monocytes, and myeloid dendritic cells, as well as the majority of acute myeloid leukemia (AML) blasts. Its extracellular region contains an N-terminal V-set immunoglobulin domain that recognizes alpha-2,3- and alpha-2,6-linked sialylated glycans, plus a C2-set domain. The cytoplasmic tail carries immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit the phosphatases SHP-1 and SHP-2, delivering inhibitory signals that dampen myeloid cell activation. Upon antibody engagement, CD33 is readily internalized, a property that underlies its use as a delivery target for cytotoxic payloads in AML. Because of its restricted myeloid expression and high prevalence on leukemic blasts, CD33 is a validated marker and therapeutic target in myeloid malignancy research.
