Background
This product is an unconjugated, non-therapeutic recombinant analog of bococizumab, an anti-PCSK9 antibody, supplied strictly for research use. It reproduces the antigen-binding specificity of the originator molecule directed against human PCSK9 (proprotein convertase subtilisin/kexin type 9), presented on a human IgG2 backbone bearing A330S/P331S substitutions in the Kappa format. Because it is manufactured as a research reagent rather than a clinical drug, it is well suited to controlled laboratory work: as a positive control or reference binder in PCSK9 binding and neutralisation assays, as a tool to probe PCSK9/LDL-receptor interactions in vitro, for ELISA and surface-plasmon-resonance characterisation, for cell-based receptor-recycling studies, and as a comparator in preclinical method development. It is offered as a bulk, low-endotoxin, RUO-grade material (research grade typically under 1 EU/mg, ultra-low under 0.5 EU/mg) available in milligram to gram quantities and in-vivo-standard preparations. PCSK9 (proprotein convertase subtilisin/kexin type 9; UniProt Q8NBP7) is a secreted serine protease of the proprotein convertase family that is a central regulator of plasma LDL cholesterol. Synthesised mainly in hepatocytes, it undergoes autocatalytic cleavage in the endoplasmic reticulum, after which the prodomain remains non-covalently bound and the mature enzyme is secreted. Rather than acting through its catalytic activity, circulating PCSK9 binds the EGF-A domain of the LDL receptor (LDLR) at the hepatocyte surface. This binding redirects the internalised PCSK9-LDLR complex toward lysosomal degradation instead of receptor recycling, lowering surface LDLR density and raising plasma LDL-C. Gain-of-function mutations cause autosomal dominant hypercholesterolaemia, while loss-of-function variants reduce LDL-C and cardiovascular risk, validating PCSK9 as a therapeutic target. Antibodies that block the PCSK9-LDLR interface preserve LDL receptors and increase LDL clearance.
