This is clone 55R-593, an Armenian Hamster IgG1 monoclonal antibody raised against mouse CD120a (TNF Receptor I, TNFR1/p55). CD120a, also known as TNF Receptor 1 (TNFR1, p55/p60, gene Tnfrsf1a; UniProt P25118), is one of the two principal receptors for tumor necrosis factor (TNF), the other being TNFR2/CD120b. TNFR1 is expressed broadly across most nucleated cell types and can be engaged by both soluble and membrane-bound TNF. It contains an intracellular death domain, and ligand engagement drives assembly of signaling complexes that can activate NF-kB and MAPK pathways to promote inflammation and cell survival, or, alternatively, recruit death-domain adaptors to trigger apoptosis or necroptosis. Through these outputs TNFR1 is a central mediator of inflammatory responses, host defense, tissue homeostasis, and, when dysregulated, of chronic inflammatory and autoimmune pathology. Because it drives much of TNF's pro-inflammatory and cytotoxic activity, TNFR1 is a key node studied in models of sepsis, autoimmunity, and inflammatory tissue injury.
