Background
ICH5139 is an unconjugated, non-therapeutic recombinant analog of abrilumab (AMG 181 / MEDI 7183), a fully human IgG2 kappa monoclonal antibody directed against the alpha4-beta7 integrin heterodimer. Built around the same target specificity as the originator drug, this research-grade biosimilar is intended solely for research use (RUO) and is not the clinical product. It gives investigators a reliable tool for studying alpha4-beta7-dependent leukocyte adhesion and gut-selective lymphocyte trafficking without sourcing scarce clinical material. Typical applications include use as a positive control or benchmark antibody in binding and neutralisation assays, in-vitro cell-adhesion and blocking studies, epitope and competition work, assay development and standardisation, and preclinical model characterisation. It is supplied unconjugated, low-endotoxin (research grade less than 1 EU/mg; ultra-low grade less than 0.5 EU/mg), and available in bulk milligram-to-gram quantities to support scale-up and reproducibility. Not for human or veterinary use. The molecular target is the alpha4-beta7 integrin, a heterodimer of integrin subunit alpha-4 (ITGA4, P13612) paired with integrin subunit beta-7 (ITGB7, P26010). Note that ITGA4 can also pair with beta-1 to form alpha4-beta1 (VLA-4); alpha4-beta7 specifically confers gut-selective homing. Expressed on subsets of memory T and B lymphocytes, alpha4-beta7 binds mucosal addressin cell-adhesion molecule-1 (MAdCAM-1) on gut endothelial venules, and also fibronectin and VCAM-1. This interaction mediates the rolling, firm adhesion, and extravasation of leukocytes into gut-associated lymphoid tissue and intestinal mucosa. Because MAdCAM-1 expression is largely restricted to the gastrointestinal tract, selectively blocking alpha4-beta7 reduces immune-cell recruitment to the gut while sparing systemic and central-nervous-system trafficking, the basis of gut-selective anti-integrin strategies in inflammatory bowel disease research.
