Background
This product is an unconjugated, non-therapeutic recombinant analog of clazakizumab, a humanized monoclonal antibody directed against interleukin-6 (IL-6). It is supplied as a research-use-only (RUO) reagent and is not the clinical drug, nor is it intended for human or veterinary use. Built as a human IgG1 that binds the IL-6 cytokine, it is intended to recapitulate the antigen-binding behaviour of the originator so researchers can study IL-6 neutralisation in controlled systems. Typical applications include in-vitro blockade of IL-6 signalling, use as a positive/reference control in binding and neutralisation assays, epitope and affinity characterisation, and as a benchmark antibody in biosimilarity or developability workflows. Manufactured to research grade with low endotoxin (<1 EU/mg standard; ultra-low <0.5 EU/mg options) and available in bulk milligram-to-gram quantities, it supports assay development, in-vitro mechanistic studies, and preclinical formatting work. Note that this is an anti-cytokine biosimilar analog, distinct from anti-IL-6-receptor antibodies such as tocilizumab, which target the receptor rather than the ligand. Interleukin-6 (IL-6; UniProt P05231) is a pleiotropic ~21-26 kDa four-helix-bundle cytokine central to inflammation, acute-phase responses, and immune regulation. It is produced by many cell types, including macrophages, T cells, fibroblasts, and endothelial cells. IL-6 signals through two modes: classic signalling, in which it binds the membrane IL-6 receptor (IL-6R/CD126) on hepatocytes and leukocytes; and trans-signalling, in which IL-6 complexed with soluble IL-6R engages the ubiquitously expressed signal-transducing subunit gp130 (IL6ST). Both routes drive gp130 dimerisation and activation of the JAK/STAT3, SHP2/MAPK, and PI3K pathways. Downstream effects include hepatic C-reactive protein production, B-cell and plasma-cell differentiation, Th17 polarisation, and osteoclast activity. Dysregulated IL-6 contributes to rheumatoid arthritis, cytokine release syndrome, and other inflammatory conditions, making the ligand a well-validated therapeutic and research target.

Purification Method
