This product is an unconjugated, non-therapeutic recombinant analog of pacmilimab (CX-072), an investigational anti-PD-L1 antibody, supplied strictly for research use only. It is a human IgG4 (kappa) built around the PD-L1 target to reproduce the antigen-binding behavior of the originator, and it is not the clinical drug and is not intended for human or veterinary use. The originator is notable as a conditionally activated "Probody" prodrug in which a masking peptide restricts binding until tumor-associated proteases remove it; this research-grade analog is offered as a standard unmasked antibody format for straightforward benchtop use unless otherwise specified. It is useful as a benchmarking reagent and positive/isotype-matched control in PD-L1 binding, blockade, and checkpoint-assay development, for comparison against other anti-PD-L1 analogs, and for antibody-engineering, ADCC/ADC-development, and preclinical model work. Available at low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and in bulk mg-to-gram quantities to support screening, assay standardization, and scale-dependent studies. Guidance for ichorbio to review. PD-L1 (Programmed Death-Ligand 1; also CD274 or B7-H1; UniProt Q9NZQ7) is a type I transmembrane glycoprotein of the B7 immunoglobulin superfamily. It is expressed on many tumor cells, antigen-presenting cells, and various stromal and epithelial tissues, and is upregulated by inflammatory signals such as interferon-gamma. PD-L1 binds the inhibitory receptor PD-1 (PDCD1) on activated T cells, delivering a co-inhibitory signal that dampens T-cell receptor signaling, cytokine production, and effector function. It also engages CD80 (B7-1). Through these interactions PD-L1 contributes to peripheral immune tolerance and limits chronic inflammation, but tumors co-opt this axis to evade immune attack. Blocking PD-L1's engagement of PD-1 releases this brake and can restore anti-tumor T-cell activity, which is the rationale underlying PD-L1-directed checkpoint agents.
