IL31RA (interleukin-31 receptor subunit alpha; UniProt Q8NI17) is a type I cytokine receptor chain and the ligand-specific component of the interleukin-31 receptor. It does not signal alone: IL-31 first engages IL31RA, and the complex then recruits the oncostatin M receptor beta chain (OSMR) to assemble the functional heterodimeric receptor. Receptor assembly activates JAK1/JAK2 and downstream STAT3 (with lesser STAT1 and STAT5 input), as well as PI3K/AKT and MAPK branches. IL31RA is expressed on sensory dorsal root ganglion neurons, keratinocytes, monocytes/macrophages, and epithelial cells, positioning it at the interface of the immune system, epithelium, and the peripheral nervous system. This axis is a principal driver of pruritus and is implicated in type 2 inflammatory skin disease, making IL31RA a well-studied target for itch and inflammation research.
