IL6R (UniProt P08887) is the specific, non-signaling alpha subunit of the interleukin-6 receptor. Interleukin-6 first binds IL6R, and this IL-6/IL6R complex then recruits and dimerizes the signal-transducing beta subunit gp130 (IL6ST), triggering JAK/STAT3 signaling that drives acute-phase responses, immune-cell differentiation, and inflammation. IL6R exists as a membrane-bound form on hepatocytes and leukocytes ("classic" signaling) and as a soluble form (sIL6R) that enables "trans-signaling" on cells lacking membrane IL6R but expressing gp130, broadening the range of responsive cell types.
