Background
This product is an unconjugated, non-therapeutic recombinant analog of the originator anti-Factor XI antibody gruticibart, supplied for research use only. It is a full-length human IgG4-kappa immunoglobulin built around the coagulation Factor XI (F11, UniProt P03951) target, produced recombinantly and intended as a laboratory reagent rather than the clinical drug; it is not for human or veterinary use. The analog is useful for researchers studying the contact and intrinsic coagulation pathways, FXI/FXIa biology, and antibody-based anticoagulation mechanisms. Typical research applications include use as a binding and neutralization reference standard, a benchmarking control in FXI target-engagement and epitope-competition assays, a tool for characterizing FXI activation by Factor XIIa, and a comparator in preclinical assay development and biosimilarity workflows. Each subunit contains four apple domains (A1-A4) and a C-terminal serine protease domain. FXI is classically activated to FXIa by Factor XIIa during contact activation, and can also be activated by thrombin; the apple 2 (A2) domain participates in FXIIa-mediated activation. Activated FXIa cleaves Factor IX to propagate thrombin generation and amplify clot formation. Because FXI contributes more to pathological thrombosis than to normal hemostasis, it is a leading target for anticoagulants aiming to separate antithrombotic efficacy from bleeding risk. Loss-of-function reflects hemophilia C, generally a milder bleeding phenotype, underscoring the therapeutic rationale for FXI inhibition.
