Background
ICH5076 is a research-grade, unconjugated recombinant analog of sacituzumab, a human IgG1 antibody directed against TACSTD2 (Trop-2). It is supplied as a non-therapeutic reagent for research use only and is not the clinical product; it is intended for laboratory and preclinical applications rather than human or veterinary use. Because it is built around the Trop-2 target and carries a human IgG1 framework, it is well suited to in-vitro binding characterization, epitope and cross-reactivity studies, flow cytometry of Trop-2-expressing cells, and as a targeting moiety in antibody-drug conjugate (ADC) development workflows where sacituzumab is the reference antibody. Being unconjugated, it also serves as a naked-antibody control against conjugated formats. The material is offered at low endotoxin (research grade typically <1 EU/mg, with ultra-low <0.5 EU/mg options) and in bulk milligram-to-gram quantities, supporting assay standardization, positive/negative controls, and preclinical model work. Investigators should validate lot performance in their own systems before use.
TACSTD2 (tumor-associated calcium signal transducer 2), commonly called Trop-2, is a single-pass type I transmembrane glycoprotein encoded by an intronless gene. Its extracellular region contains a cysteine-rich domain and a thyroglobulin type-1 repeat, followed by a transmembrane segment and a short cytoplasmic tail bearing a PIP2-binding sequence and a conserved serine phosphorylation site. Trop-2 participates in intracellular calcium signaling and modulates proliferation, adhesion, and migration, in part through regulated intramembrane proteolysis that releases signaling fragments. It is expressed in many normal epithelia but is markedly overexpressed across numerous carcinomas, including triple-negative breast, urothelial, and lung cancers, where high expression often correlates with aggressive behavior. This tumor-associated overexpression and cell-surface accessibility make Trop-2 an attractive target for antibody-based and ADC therapeutics, and a useful antigen for research reagents.

Purification Method
