Background: Tumor-associated macrophages (TAMs) are notoriously difficult to deplete and accurately quantify in the tumor microenvironment. Standard depletion protocols utilizing Anti-CSF1R antibodies can complicate downstream flow cytometry analysis if the validation clones compete for the same receptor space. To guarantee accurate immune profiling, ichorbio has engineered a dual-target Macrophage Depletion & Validation Kit. We utilize our low endotoxin Anti-Mouse CSF1R (Clone AFS98) to drive the in vivo depletion by blocking the colony-stimulating factor 1 receptor. For validation, the kit includes Clone CI:A3-1, which targets the F4/80 macrophage surface protein. By targeting two entirely different structural markers on the macrophage, this kit provides a foolproof, masking-free readout of your in vivo depletion efficacy. These monoclonal antibodies were purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype.

Depletion antibody: Phosphate buffered saline (PBS) pH 7.2,with no carrier protein, potassium or preservatives added. BSA and Azide free. Validation Antibody: Phosphate buffered solution, pH 7.2, containing 0.09% stabilizer.

Depletion antibody: Can be frozen. Validation antibody: can be stored at 2-8°C for 24 months. Please protected from prolonged exposure to light and do not freeze.
