NKG2A (encoded by KLRC1; UniProt P26715) is an inhibitory C-type lectin-like receptor expressed on natural killer cells and subsets of CD8 T cells. It heterodimerizes with CD94 (KLRD1) to form the CD94/NKG2A complex, which recognizes the non-classical MHC class I molecule HLA-E presenting leader peptides derived from other MHC class I molecules. Engagement delivers an inhibitory signal through cytoplasmic ITIM motifs that recruit the phosphatases SHP-1 and SHP-2, dampening cytotoxicity and cytokine release and contributing to self-tolerance and NK "missing-self" surveillance.
