Background
This is an unconjugated, non-therapeutic recombinant analog of Imelciment (originator code REGN-9035), supplied for research use only and not for human or veterinary use. Notably, Imelciment is itself an anti-idiotype / reversal agent: its target is not a conventional cell-surface antigen but the investigational NPR1-agonist antibody vixticibart (REGN5381). The analog is therefore built to recapitulate binding to vixticibart, letting laboratories reproduce the drug/reversal-agent pairing in vitro. It is presented as a half-IgG4-kappa (h-CH2-CH3) format consistent with an engineered anti-idiotype binder. Because it is a defined recombinant reagent, it is useful as an assay control, a capture/detection tool for vixticibart, and a neutralizing counter-reagent in binding and functional studies. It is offered at research grade with low endotoxin (typically <1 EU/mg research grade, <0.5 EU/mg ultra-low) and can be supplied in bulk milligram-to-gram quantities and as an in-vivo-standard-grade preparation for preclinical modeling. It is expressly not the clinical drug and carries no therapeutic claims. The nominal target here is vixticibart (REGN5381), an investigational human IgG4-kappa monoclonal antibody that acts as an allosteric agonist of natriuretic peptide receptor 1 (NPR1, also called NPR-A / GC-A). NPR1 is a single-pass, membrane-bound receptor guanylyl cyclase that is physiologically activated by atrial and B-type natriuretic peptides (ANP, BNP) to raise intracellular cGMP, driving vasodilation, natriuresis and diuresis. Vixticibart binds the NPR1 extracellular domain and stabilizes an active-like conformation, increasing cGMP output independently of, and additively with, endogenous peptides; it is selective for NPR1 over NPR2 and NPR3. Imelciment binds vixticibart itself, functioning as an anti-idiotype reversal agent intended to neutralize the agonist's activity. Understanding NPR1 signaling is thus central to interpreting how this reagent pair behaves.
